Verification of pig liver TTR gene promoter on transgenic mouse living body level

Abstract

The invention belongs to the technical field of animal gene engineering, and particularly relates to application of a pig liver TTR gene promoter in transgenic mouse genetic manipulation. On the basis of earlier research, a pTTRs-BGLhis-control vector is subjected to linearization treatment, the linearization segment is subjected to protocaryon microinjection to obtain a transgenic mouse, and the liver tissue specificity of the promoter on the living body level is verified. The result indicates that the promoter can be expressed in the specific driven exogenous gene BGL1 in the mouse liver tissue and generate beta-glucosaccharase with certain enzyme activity. The pig TTR gene promoter is applied to the transgenic animal, thereby providing an available tissue-specific promoter for animal gene engineering, providing conditions for enhancing the digestion utilization ratio of the nonruminant animal feed and providing important functional elements for improving productivity of livestock and poultry by gene engineering technology.
