Abstract

The present invention pertains to the field of biotechnology, in particular to a tumor necrosis factor-related apoptosis-inducing ligand variant. The tumor necrosis factor-related apoptosis-inducing ligand variant is a fusion protein which consists of integrin ligands of AlphaVBeta3 and AlphaVBeta5, a linking peptide and a tumor necrosis factor-related apoptosis-inducing ligand, and the present invention is produced by the construction of an encoding gene of the variant by a genetic engineering method, the soluble recombinant expression and the simple separation and purification. The tumor necrosis factor-related apoptosis-inducing ligand variant has good tumor tissue targeting property and significantly enhanced anti-tumor effect, which can also reduce the dose of the needed protein for the target treatment effect, improve the bioavailability, reduce the treatment cost, decrease and overcome the potential toxic and side effects of the tumor necrosis factor-related apoptosis-inducing ligand. The present invention further provides an expression method for the production of the tumor necrosis factor-related apoptosis-inducing ligand variant which is soluble and has high content of the polymer, and a simple separation and purification process.
