Abstract
PROBLEM TO BE SOLVED: To provide engineered lung tissue usable for lung transplantation which still remains the only definitive treatment of several lung diseases, including cystic fibrosis, emphysema/COPD, and idiopathic pulmonary fibrosis and also for in vitro three-dimensional lung culture models which can be used to study a wide variety of important aspects of pulmonary biology and physiology.SOLUTION: The invention relates to a decellularized tissue capable of supporting cell growth and also exhibits a characteristic of a corresponding natural tissue prior to decellularization. The decellularized tissue retains an extracellular matrix of the corresponding natural tissue, wherein the extracellular matrix comprises a substantially intact exterior surface. Immunogenic markers have been substantially removed from the decellularized tissue. The decellularized tissue exhibits mechanical properties substantially similar to that of a corresponding natural tissue. The decellularized tissue is capable of supporting and maintaining the differentiation state of a lung cell. A population of cells comprises a stem cell, and epithelial and endothelial cells. The cells are genetically modified.
