Abstract

PROBLEM TO BE SOLVED: To provide engineered lung tissue that can be used for lung transplantation which remains the only definitive treatment for several lung diseases including cystic fibrosis, emphysema/COPD, and idiopathic pulmonary fibrosis, and that can be used for in vitro three-dimensional lung culture models which can be used for a wide range of variety of important studies of pulmonary biology and physiology.SOLUTION: A decellularized tissue is capable of supporting cell growth, and exhibits a characteristic of a corresponding natural tissue prior to decellularization. A decellularized tissue retains an extracellular matrix of a corresponding natural tissue, where: the extracellular matrix comprises an exterior surface that is substantially intact; immunogenic markers have been substantially removed; the tissue exhibits a mechanical property substantially similar to that of the corresponding natural tissue; the tissue is capable of supporting and maintaining the differentiation state of a lung cell; the tissue exhibits an intact airway tree and vascular network; the population comprises a stem cell, an epithelial cell and an endothelial cell; and the cells are genetically modified.SELECTED DRAWING: None
