Abstract

The invention discloses a tissue engineering device used for light-operated insulin rhythmic secretion. It is helpful that the tissue engineering device can be used for treating diabetes instead of insulin injection. A construction method comprises following steps: a virus expression vector of VP16-CRY2-IRES-Ga14BD-CIB1N obtained via expression driven by Ga14-UAS is constructed, and an insulin virus expression vector obtained via expression driven by Ga14/UAS is constructed; transfection of the virus expression vector and transfection of the insulin virus expression vector to skin-source fibroblast are carried out respectively via virus transfection or other methods; tissue engineering scaffolds are loaded with the modified fibroblast further, and then are subjected to subcutaneous transplantation; after stabilization of regenerous tissues in vivo, local skin is subjected to blue light irradiation so as to induce interaction of VP16-CRY2 with Ga14BD-CIB1N, induce increasing of self expression further, and form positive feedback, and at the same time, promote insulin secretion on another carrier, wherein when blue light irradiation is stopped, a system is turned into quiescent conditions, and insulin secretion is stopped. It is expected that the tissue engineering device is capable of realizing demand-based insulin secretion.
