23 Jul Tissue-engineered epidermis and preparation method thereof
Abstract
The invention belongs to the field of histocytology and tissue engineering and discloses a tissue-engineered epidermis with hypodermal cells and a preparation method thereof. The method for preparing the tissue-engineered epidermis with hypodermal cells comprises the following steps: obtaining epidermis cells; inoculating the obtained epidermis cells, culturing the cells until the density is 80-100 percent by using an epidermis culture medium, replacing the medium into a mixed culture medium, culturing for 16-24 hours, and collecting the epidermis cell layer, thereby obtaining the tissue-engineered epidermis piece; transferring the obtained tissue-engineered epidermis onto a semi-permeable polymer or a silicone mold substrate or a petrolatum gauze, thereby obtaining a film with the epidermis cell layer; and culturing the hypodermal cells, paving the obtained hypodermal cells on the epidermis cell layer with the film of the epidermis cell layer, and culturing for 1-2 hours, thereby obtaining the product. The preparation method provided by the invention is easy to operate and short in period, the tissue-engineered epidermis can be mutually stimulated and induced with receptor skin tissues, and formation of a complete epidermal structure of the receptor is promoted.
