Abstract
The invention discloses a three-dimensional liver-like tissue model construction method based on organs-on-chips. The method mainly comprises the following steps: preparing and modifying the organs-on-chips; in-situ formation of an embryoid body EBs; differentiation and maturation of liver-like organoids in the organs-on-chips. The novel engineered three-dimensional liver-like development model isa perfusion microreactor based on an organs-on-chips technology. According to the invention, the basic principle of in-vivo liver development is combined with an engineering technology, so that the microenvironment formed by the liver is better simulated. The method has the advantages of being low in cost, easy to operate, capable of achieving in-situ tracking and real-time monitoring and the like, can replace an animal model and a traditional two-dimensional culture mode, simulates formation and development of the liver to a certain extent, and provides powerful technical support for in-vitro simulation of liver formation, drug metabolism, drug screening, toxicity detection and the like.
