Abstract

The embodiment of the invention provides a serum-containing corneal epithelial cell nutrient solution, and relates to the field of biomedicine. A nutrient solution 1 is prepared from a basal culture medium, 10 to 20 percent fetal calf serum, 1 to 10 mu M retinoic acid serving as a cell proliferation and differentiation regulatory factor, and 1 to 10 mu M glycolic acid serving as a cell proliferation and differentiation regulatory factor; a nutrient solution 2 is prepared from a basal culture medium, 1 to 5 percent fetal calf serum, 1 to 5 mu M retinoic acid serving as a cell proliferation and differentiation regulatory factor, 1 to 5 mu M glycolic acid serving as a cell proliferation and differentiation regulatory factor, 1 to 5 mu M Rock inhibitor Y27632, 1 to 5 mu M CACl2, 5 to 15 mu g/ml insulin, 5 to 15 mu g/ml transferrin, 5 to 20ng/ml epidermal growth factor (EGF), 5 to 20ng/ml insulin growth factor (IGF-I), 5 to 20ng/ml transforming growth factor (TGFbeta), 5 to 20ng/ml bovine pituitary extract (BPE) and 5 to 20ng/ml anchoring factor fibronectin. By adopting the serum-containing corneal epithelial cell nutrient solution, the demand of high-density proliferation of primary corneal epithelial cells can be met, and the phenomenon of inhibition of cell proliferation caused by over-differentiation in serial sub-cultivation of the epithelial cells is avoided, the proliferation and differentiation speeds of in-vitro corneal epithelial cells are balanced effectively, and the doubling time of the cells is shortened, so that the demand on building of a large-scale tissue engineering model can be met.
