23 Jul Self-assembled hydrogel based on specific interaction and preparation method thereof
Abstract
The invention discloses a self-assembled hydrogel based on specific interaction. A preparation method for the self-assembled hydrogel comprises the following steps: grafting micromolecular methacrylicanhydride onto a hyaluronic acid (HA) molecule to introduce a double bond; then grafting a polypeptide (knob) with a sequence of GPRPAAC onto the HA molecule via a click reaction of a thiol-double bond so as to obtain knob-g-HA; and allowing the knob on a knob-g-HA molecule and a fibrinogen molecule to undergo knob-hole specific interaction so as to obtain the in-situ self-assembled hydrogel. Thehydrogel prepared in the invention is of a porous structure with interlaced penetration performance and has certain mechanical properties; and the biocompatibility of the hydrogel is better comparedwith hydrogels prepared through conventional UV cross-linking preparation methods. The self-assembled hydrogel is prepared in a physiological environment, is simple and convenient to prepare and doesnot need extra crosslinking agent and the like; and the novel, gentle and rapid method is provided for preparation of in-situ self-assembled hydrogel with good biocompatibility, and has great potential in tissue engineering and regenerative medicine.
