Abstract
The invention discloses a screening and identifying method for a corn endosperm specific promoter in the early stage of grouting. The method comprises the steps that a gene promoter which is efficiently expressed in the corn endosperm in the early stage of grouting is screened and cloned; the expression characteristics of candidate genes are identified, a target gene betl2 is screened out, and a promoter pbetl2 of the target gene is cloned; a transient expression vector is built; different tissue of corn is bombarded through particle bombardment, and the condition that the pbetl2 drives a GUS gene to be expressed is analyzed; the condition that the promoter controls a GFP gene to be expressed in leaves is observed through corn leaf protoplast transformation; the expression conditions of the promoter in the early stage, the middle stage and the late stage of grouting are analyzed. According to the method, the specific promoter pbetl2 in the early stage of corn grouting is cloned, the important theoretical significance of artificial modification of the metabolic activity in the early stage of grouting on research of endosperm development related mechanism is achieved, and meanwhile the important practical significance is achieved on specific gene expression in the early stage of corn grouting through a genetic engineering technology.
