Abstract

Belonging to the technical field of biological engineering, the invention particularly relates to recombination of an ER (estrogen receptor) beta promoter luciferase reporter gene, construction of an estrogen receptor beta promoter cell model, and application in screening estrogen replacement drugs for effectively mitigating periodontal tissue diseases and maintaining the alveolar bone height. In the invention, the genetic engineering technology is employed to amplify a 1253bp ER beta promoter sequence, which is cloned to a PGL2-luc carrier to construct an ER beta promoter-Luc expression vector. The ER beta expression is controlled by a promoter region. Construction of a periodontal ligament cell ER beta promoter model is a key step in screening of effective Chinese medicinal components treating bone defect. Construction of the drug screening cell model proves that the American ginseng component of certain concentration can promote ER beta promoter transcription, and the higher the concentration is, the stronger the activity is, thus guiding clinical application of the drug in treatment of periodontal diseases and alveolar bone resorption.
