Abstract
The invention discloses a rat tail intervertebral disc in-vitro three-dimensional gel stereoscopic culture method which comprises the following steps: preparing a 2 wt% sepharose gel with a PBS (phosphate buffer solution), and mixing with a DMEM (dulbecco’s modified eagle medium) culture solution containing 10 wt% of FBS (fetal bovine serum) in a volume ratio of 1:1 to form a three-dimensional culture gel; immediately disinfecting a rat tail cut from an in-vivo C57 line rat, removing the skin, and cleaning in the PBS; under a stereoscopic microscope, removing the ligament near the vertebrae to reveal the intervertebral disc, cutting off the complete intervertebral disc along the upper and lower cartilage endplates, and cleaning with the PBS; and spreading the three-dimensional culture gel on a culture dish, quickly transplanting the intervertebral disc into the three-dimensional culture gel, solidifying, adding the DMEM culture solution containing 10 wt% of FBS to cover the surface, putting in a culture box, and changing the DMEM culture solution containing 10 wt% of FBS once a day. The method is suitable for researches of intervertebral disc tissue engineering science, intervertebral disc transplantation, pharmaceutical control on intervertebral disc degradation and the like.
