Abstract
The invention relates to a protease K and application thereof. The protease K has a nucleotide sequence of SEQ ID No. 2 and an amino acid sequence of SEQ ID No. 1. The protease K screened in the invention cuts carboxyl-terminal peptide bonds of aliphatic amino acids and aromatic amino acids, can be used for preparing chromosome DNA for pulse electrophoresis, western blotting and removal of nuclease in preparation DNA and RNA, can also be used for extraction of non-protein components in tissue for degradation of foreign matters containing protein, e.g., DNA vaccine and heparin preparation, and has an industrial production application value. A screened protease K gene can be used for transformation of engineering bacteria so as to obtain recombinant bacteria expressing protease K.
