23 Jul Promoter of serine carboxypeptidase-like protein gene AtSCPL30 of Arabidopis thaliana and deletion mutants thereof, and application of promoter and deletion mutants
Abstract
The invention discloses a promoter of the serine carboxypeptidase-like protein gene AtSCPL30 of Arabidopis thaliana and deletion mutants thereof. The promoter is named as PD1 and is a regulatory sequence with a length of 2093 bp from the upper stream of the translation initiation codon ATG of the serine carboxypeptidase-like protein gene AtSCPL30 of Arabidopis thaliana; and the nucleotide sequenceof the promoter is as shown in SEQ ID No. 1. The deletion mutants are eight continuous sequence segments obtained after deletion of segments of different lengths from the 5′ terminal of the sequenceof the PD1, and the eight continuous sequence segments are sequentially named as PD2 to PD9 and have corresponding nucleotide sequences as shown in SEQ ID No. 2 to SEQ ID No. 9. The invention also discloses application of the promoter and the deletion mutants thereof to plant functional gene research or plant genetic engineering breeding. Experimental results prove that PD1 to PD9 can promote thehigh-intensity expression of downstream gene (such as GUS reporter genes) in each tissue of transgenic plant receptors, and are almost insusceptible to induction or inhibition by high-salt stress, osmotic stress and adversity stress like low temperature and low phosphorus. The PD1 to PD9 are constitutive strong promoters and have immense application value.
