Preparation of core-shell structure composite material as well as method for constructing tissue engineering microstructure by using core-shell structure composite material

Abstract

The invention provides preparation of a core-shell structure composite material as well as a method for constructing a tissue engineering microstructure by using the core-shell structure composite material. The method concretely comprises the following steps: sequentially decalcifying, defatting and deproteinizing limb bones of purchased and freshly slaughtered large mammals, and then crushing toobtain bone particles; then, defatting, decalcifying and deproteinizing the obtained bone particles again to obtain decalcified bone matrix (DBM) microparticles; adding gelatin powder into a phosphatebuffer solution (PBS) to prepare a gelatin solution, then applying the gelatin solution into micropores formed in a polydimethylsiloxane (PDMS) plate, adding the 2-5 DBM microparticles into each micropore, cross-linking, rinsing, and freeze-drying to obtain the core-shell structure composite material; pushing the obtained composite material out by using an ejector pin, then agglomerating and placing in a petri dish, and dropwise adding prepared cell suspension into the core-shell structure composite material for culturing so as to obtain the tissue engineering microstructure. The core-shell structure composite material prepared by the method not only has good biocompatibility of the gelatin, but also has good mechanical properties of a DBM, so that blood vessels can better grow into the composite material after the composite material is implanted into a human body.
