Abstract
The invention discloses a preparation method of tissue engineering adipose-derived stem cell lamella being used for cartilage injury repair and having cartilage differentiation potential. In the method, by preparing a specific culture system, the ASCs lamella, produced through a cell lamella culture technology, has a complete sheet structure containing extracellular matrix and has cartilage differentiation potential. Because only is the tissue, formed by pure cells, transplanted into body, use of supporters, like a scaffold, and adverse reactions due to the supporters are avoided. The extracellular matrix remains on the ASCs lamella, so that the ASCs lamella contains important cell surface proteins, such as ion channels, growth factor receptors and connexins, etc.; coordination of intercellular functions can be maintained, so that the method has unparalleled advantages than a conventional tissue engineering technology. The complete lamella structure is beneficial to retention of the cells at the transplanting position, so that utilization rate of the cells and activity of the transplanted cells are greatly improved.
