Abstract

The invention provides a preparation method of an in-vitro vascularization 3D tissue based on a micromachining technology, which belongs to the fields of microfluidic technologies, tissue engineering,material chemistry and the like. The method comprises the steps of soft template preparation, endothelial tissue pre-forming, in-vitro 3D tissue block forming, in-vitro 3D tissue block culture and the like. According to the invention, a template is manufactured by using an accurate and controllable micromachining technology; methacrylated gelatin with excellent biocompatibility and processabilityand polyethylene glycol diacrylate are used as scaffold materials to prepare co-culture tissue blocks of endothelial cells and other tissue cells, and the method has great application value in the aspects of vascularization in-vitro micro-tissue construction, tissue physiology and pathology research and the like.
