Preparation method of in-situ cross-linked hyaluronic acid hydrogel

Abstract

The invention relates to a preparation method of in-situ cross-linked hyaluronic acid hydrogel and belongs to the technical field of biomedical materials. According to the preparation method, partialring-opening oxidization is carried out on hyaluronic acid by adopting sodium periodate, and hydroxyacetic acid is grafted with polyvinyl alcohol; then in-situ gelled interpenetrating double-network hydrogel is prepared through Schiff base reaction and sulfydryl oxidization; the hydrogel has high gel strength, high fracture toughness and fracture stress, and a gel material with high mechanical strength can be formed; a low molecular weight crosslinking agent with a cytotoxic effect is successfully avoided and a catalyst is also not needed; the preparation method is simple and convenient to operate; compared with typical natural high molecular hydrogel, the mechanical property and the anti-degrading capability are improved and a caused inflammatory effect is relatively low; the hydrogel haslong retention time, low cytotoxic effect and good biocompatibility; a hydrogel system of crosslinking reaction can be finished in bodies, without the triggering of any special condition, so that a novel tissue engineering scaffold capable of administrating a drug to form gel is provided for tissue regeneration.
