Abstract
The invention discloses a preparation method of all-organ acellular matrix of a heart. In the method, perfusion with 1% SDS solution and 1% Ttiton is performed, then PBS is used for flushing, liquid is exchanged for 4-5 times every day, and cleaning is totally performed for 72-128h. According to the method, a reasonable all-organ acellular system is designed, an existing heart all-organ acellular method is improved and optimized, and acellular treatment is performed according to different eluting sequences and time proportions by combining with SDS detergent and TritonX100, so that the eluting effect of the perfusate in the heart organ is improved, and a foundation is built for application of the method in elevation of late acellular matrix and reconstruction of tissue engineering. The all-organ acellular matrix of the heart prepared by the method can reserve original second-level and third-level vessel structures, supermicro conformation and extracellular matrix components of the heart under the condition of removing original resident cells of the original tissue.
