Abstract
The invention relates to the technical field of biomedical engineering, in particular to a preparation method and application of a tissue engineering material for promoting directional differentiation of mesenchymal stem cells. The preparation method provided by the invention comprises the following steps: transplanting the mesenchymal stem cells to a PLGA (poly(lactic-co-glycolic acid)) stent, by controlling the time, concentration and mode of planting the mesenchymal stem cells as well as the size, the shape and the like of a stent material, searching out optimum conditions for the directional differentiation of the mesenchymal stem cells, and observing biocompatibility of the mesenchymal stem cells and the PLGA stent, change in cellular morphology during inducing, influence of a pancreatic beta cell inducing liquid on cell proliferation as well as acquisition of specific genes, proteins and functions inducing cell differentiation. According to the preparation method disclosed by the invention, the mesenchymal stem cells are planted on the PLGA stent under the optimum conditions, and a pancreatic beta cell, which has high glucose reactivity and is applicable to therapy transplantation, is obtained from culture and differentiation by virtue of the pancreatic beta cell inducing liquid; therefore, the preparation method and application bring hope to the treatment of diabetes.
