Abstract
The perfusion bioreactor for the production and/or cultivation of human or animal tissues or tissue equivalents, where the production is based on a construct cultivated in the interior, comprises a cover (1), an inlet (2) and outlet (3) for a fluid nutrient medium. The inlet is connected with a unit for producing a perfusion pressure. A part segment of the cover consists of elastic material. The elastic part material corresponds to more than 75% of the interior surface of the cover surface. The bioreactor consists of reabsorbing or not reabsorbing porous skeleton. The perfusion bioreactor for the production and/or cultivation of human or animal tissues or tissue equivalents, where the production is based on a construct cultivated in the interior, comprises a cover (1), an inlet (2) and outlet (3) for a fluid nutrient medium. The inlet is connected with a unit for producing a perfusion pressure. A part segment of the cover consists of elastic material. The elastic part material corresponds to more than 75% of the interior surface of the cover surface. The bioreactor consists of reabsorbing or not reabsorbing porous skeleton, guiding system and hollow fiber system for the distribution of the culture medium in the tissue to be produced or tissue equivalent. The construct subjected in the interior area is flow throughable by the nutrient medium. The interior contour of the interior area corresponds to 100% of the external contour of the tissue to be produced or tissue equivalents and the production is carried out in a given spatial configuration. The bioreactor contains a scaffold. The bioreactor is operated as a unit in connection with a self-regulatory pulsating perfusion system. The physical pressure regime, which is producible in the interior area corresponds to the physical pressure regime, which influences under normal or pathological conditions in the living human or animal organism on the produced tissues, tissue equivalents, blood vessels or blood vessel networks. The elasticity of the material of the part segment is adjusted so that the extension of the tissue and/or tissue equivalents together with the flexibility of the cover through the perfusion pressure produced in the interior corresponds to the physiological or pathological values of the tissue compliance of the tissue to be produced. Connections and devices for a monitoring are operated to begin the fabrication process. The interior area is divided into two compartments that are disposed over self through-flow circuit and are subdivided through detachable or non-detachable separation wall. A visibility disk is subjected for optical or functional monitoring directly in the elastic cover. The tissue, blood vessel, blood vessel equivalents or corresponding equivalents are produced by the bioreactor. A soft-tissue or soft tissue equivalents are produced by the bioreactor. The production of the tissue equivalents is carried out through filling the hollow area with a reabsorbing carrier material that is presettled with human or animal cells such as mesenschymal cells. Endothelial cells are additionally added to the mesenschymal cells. The particle and/or endothelial cells are admitted in a fibrin matrix. The bioreactor is applicable for the unique or multiple uses. The interior contour of the bioreactor is produced through covering of the scaffold, which is formed after the defect. Independent claims are included for: (1) a method for the production of human or animal tissues, tissue equivalent by bioreactor; (2) a method for the production of artificial blood vessel system by tissue engineering; (3) method for the production of perfusion bioreactor; and (4) a hollow fiber system.
