Abstract
The invention belongs to the technical field of mutation or genetic engineering and discloses an ornamental garlic tissue culture propagation method. The method includes: selecting bulbils, with stemdiscs, of ornamental garlic as explants; selecting ornamental garlic cloves uniform in size and free of disease and pest damages, peeling the garlic cloves, and disinfecting; cutting each disinfectedgarlic clove along the bud middle into four uniform stem tips with the stem discs, and performing inoculation in a bud induced differentiation culture medium; performing inoculation of the explants inthe induced differentiation culture medium; cutting off differentiated single buds, and performing inoculation in a rooting culture medium; slowly unscrewing a bottle cap in an incubator, adding distilled water in depth of 0.5-1cm into a culture bottle, transplanting into a sterile medium, and culturing under conditions of 4000lx, 22 DEG C and 12h/d and conditions of 0lx, 16 DEG C and 12h/d to realize robust growing. By adoption of the ornamental garlic as a material, a tissue culture fast propagation system with a short period and a high propagation coefficient is created.
