According to the oral mucosa cell culture method,a serum-free medium is utilized, the cells can realize stable passage for 4-5 generations, the multiplication capacity of the former three generations is high, a certain proportion of giant cells occur in the fourth generation, the cells of the fifth generation are aged, thus from the angle of tissue engineering, the research supports the use of the cells of the cultured third generation, the certain number of cells are met, it is ensured that the cells have the good multiplication capacity, the passage number of the mucosa epithelial cells is closely related to the number of inoculated cells,uncertain factors are few, the experimental efficiency is high, disinfection and sterilization treatment is adopted before and after an experiment, the uncertain factors of the experiment are furtherreduced, and thus the experimental efficiency is improved.
