23 Jul Novel combined mutant of tissue type plasminogen activator
Abstract
A t-PA recombined derivative that is a novel combined mutant of a tissue type plasminogen activator (called rPA for short) is obtained by: starting from reconstructing a natural molecular structure of the tissue type plasminogen activator (called t-PA for short), reconstructing by protein engineering, deleting an EGF area, a Finger area and a kringle I area in a t-PA molecular, and retaining two functional areas (the amino acids at positions 176-527 of t-PA) of Kringle II and protease with thrombolysis function; and simultaneously mutating the amino acids HRR at positions 122-124 in the retained area to be AAA. The r-PA appears substantially-enhanced fibrin excitant, prolongs half life, increases proenzyme characteristic, and raises survivability inhibiting PAI-1, while having advantages of t-PA such as thrombus singularity. Thus, the t-PA restructuring derivate-combination mutant can be new generation thrombolysis medicine with more clinical advantages.
