Abstract
The invention discloses a novel adipose-derived stem cell culture method, which is mainly applied to primary culture of adipose-derived stem cells and belongs to the technical field of cell engineering. The method comprises the following steps; upper-layer grease is removed by a centrifugal method; 0.9% normal saline is added in an isopyknic mode, adipose tissue is physically broken, and adipose stem cells and adipose cells are made to become single cells or small cell clusters; and residues can be filtered by using a 200-mesh screen to remove large-particle fat and fat cells so as to obtain the adipose-derived stem cells. Compared with the prior art, the adipose tissue is physically crushed and then filtered by the 200-mesh screen, so that adipose cells are separated from adipose stem cells. The primary adipose-derived stem cells can be directly obtained by collecting and culturing the primary adipose-derived stem cells, the preparation method is simple, the cost is low, only a smallamount of tissue needs to be consumed, and the adipose-derived stem cells cannot be damaged by collagenase digestion.
