Abstract

The invention discloses a non-tissue culture corn genetic transformation method. The non-tissue culture corn genetic transformation method comprises the following steps that S1, plasmid containing a target gene is guided into an agrobacterium competent cell; S2, a corn immature embryo is prepared; S3, the agrobacterium cell is diluted in an LB liquid culture medium, and the corn immature embryo iscompletely soaked into the LB culture medium; S4, the corn immature embryo is taken out and placed on an MS culture medium and cultured for 2-3 days under a dark condition, and then the corn immatureembryo growing well on the MS culture medium is transferred to a new MS culture medium, and the step is repeated for twice or three times; S5, the corn immature embryo is transplanted into nutrient soil, and cultured in a green house; and S6, after the corn immature embryo grows to be a seedling, a corn lamina is taken and subjected to plasmid extraction, and the plasmid is detected by using PCR(polymerase chain reaction) method. The non-tissue culture corn genetic transformation method can avoid limitation of genetypes in the tissue culture process, the transformation system is simple, convenient, efficient and large-scale, and can promote more genetic engineering breeds to be applied to production, and accordingly the development of current crop biological engineering is promoted.
