Abstract

NEW MATERIAL:A protein present in human brain and having neurotrophic activity-inhibitory effects, having the following characteristics: (1) molecular weight: ca.5000 (determined by the SDS-polyacrylamide gel electrophoresis); (2) physical appearance: white amorphous powder; (3) stable pH range: 3.0-7.7; (4) thermal stability: stable even after subjected to 37 deg.C for 20hr, stable even after heated at 100 deg.C for 5hr; (5) amino acid sequence: the entire amino acid sequence of formula I USE:Effective for the diagnoses, prevention or therapies of Alzheimer’s disease. PREPARATION:An extract from human brain tissue is either directly or, after concentration, put to combined treatment comprising ultrafiltration, ion exchange chromatography, gel filtration, high performance liquid chromatography, etc., to carry out purification and then collection, thus obtaining the objective substance. Using the cDNA (having the nucleic acid sequence of formula II) coding the present protein, the present protein can be produced in large quantities by a genetic engineering technique.
