23 Jul Molecule labelling method for specific PCR detection of genes related to chicken growth and egg-laying deseription by employing allelomorph and use thereof
Abstract
The invention relates to a method for detecting the polymorphism of chicken Ghrelin gene and evaluating the growth and egg-laying character of chicken using improved ASP (allele specificity PCR) detection technology, and its detection and diagnosis kit therefor belonging to the animal gene engineering technology field. The method comprises: extracting gene genome DNAs from chicken blood or tissue, designing primers, obtaining partial sequences of Ghrelin gene, implementing PCR products cloning sequencing, improved ASP polymorphism detection, improved ASP polymorphism and egg-laying character association analysis. The results of the detection and analysis confirm that the existence of Ghrelin allele is obviously associative to the growth and egg-laying character of chicken. The invention also discloses three pairs primer sequences for amplification of special sequences of Ghrelin gene fragment, and detection, evaluation and application of base mutation site of said gene fragment and ASP polymorphism in chicken growth and egg-laying character. The invention provides a new molecule label for chicken label auxiliary selection and fowl bredding.
