MODIFIED FACTOR VIII CDNA AND ITS USE FOR PRODUCTION OF FACTOR VIII

Abstract

PROBLEM TO BE SOLVED: To express the factor VIII in hematopoietic cell lines and specifically in platelets under the employment of a tissue-specific promoter of a glycoprotein IIb(GPIIb) in order to further improve the production yield of the factor VIII in genetic engineering. SOLUTION: The modified factor VIII cDNA is disclosed wherein the B- domain of the wild type factor cDNA has been deleted and truncated Factor IX intron 1 has been inserted in two locations of the Factor VIII cDNA and a cDNA is used as a promoter which is suitable for the expression in hematopoietic cell lines and specifically in platelets.
