Abstract
The invention discloses a modification method of a photocrosslinked silk fibroin and a preparation method of in-situ drug-loaded hydrogel of the photocrosslinked silk fibroin. The method comprises carrying out photocrosslinking modification on silk fibroin through a silane coupling agent-type modifier or a silk fibroin modifier, uniformly mixing the modified silk fibroin, a photoinitiator, a photo-curing monomer, a crosslinking agent and a drug, and preparing drug-loaded silk fibroin composite hydrogel in situ through specific light source irradiation. The method can realize covalent crosslinking of the silk fibroin, monomer and crosslinking agent in a low temperature environment, significantly improve the physical and mechanical properties of the silk fibroin composite hydrogel and adjust and control crosslinking density and biodegradability of the silk fibroin composite hydrogel system through selection of a use amount and types of the modifier, and design types and amounts of added drugs according to disease difference of patients. The photocrosslinked silk fibroin composite hydrogel has a wide application prospect in the field of tissue engineering and biomedicines.
