23 Jul Method for transfecting eukaryotic cell by using augmenter of liver regeneration (ALR) gene and application
Abstract
The invention relates to a method for transfecting an eukaryotic cell by using a human augmenter of liver regeneration (HALR), belonging to production methods of biological engineering, which comprises the following steps of: constructing pCDNA3.1(-)HALR plasmid, and culturing and screening HepG2; transfecting a hepatic tumor cell line obtained after HepG2 screening with the pcDNA3.1(-)HALR plasmid; and then screening the transfected hepatic cell line, selecting the transfected hepatic cell line with fast growth by a limiting dilution method for clone, and establishing a stable hepatic cell line. The method for transfecting the eukaryotic cell by using the human augmenter of liver regeneration (HALR) has great development potential as the biological artificial liver construction, can be mass-produced by using the gene engineering technology, reduces the cost, and is applicable to the research and clinic of the biological artificial liver. Besides, the transfected hepatic cell can possibly promote the growth and the propagation of the implanted cell in the hepatic cell implantation, or can possibly facilitate the development of tissue and organ engineering technology and the like.
