Abstract

The invention relates to a method for structuring and culturing salt-tolerant medicago sativa, which comprises the following steps: 1) selecting a medicago sativa variety capable of easily generating in vitro leaf embryonic callus; 2) selecting a culture medium capable of using in medicago sativa in vitro culture and generating callus tissue; 3) selecting an agrobacterium tumefaciens variety capable of conversing medicago sativa, 4) selecting binary plasmids of the agrobacterium tumefaciens, wherein recombinant gene is under the control of an efficient promoter; 5) cloning integral expressed genes of a flow pump, implanting into the binary plasmids of the agrobacterium tumefaciens, 6) introducing the recombinant binary plasmids in the agrobacterium tumefaciens, infecting in vitro leaf tissue of medicago sativa; and 7) selecting and culturing integral plant of transgene medicago sativa. The invention provides a novel approach for structuring and culturing the transgene salt-tolerant medicago sativa to improved alkali soil, which changes the alkali soil improvement from an earth work with large scale into a high and new technology genetic engineering. The initial input of the novel method is less, and the characteristic of biological infinite breeding enables inestimable long-term benefits.
