Abstract

The invention relates to a method for sorghum sudanense in-vitro induction of tracheid differentiation, and belongs to the technical field of plant cell engineering. The method comprises the steps: taking a light yellow granular callus tissue cultured and induced by a sorghum sudanense seed tissue as an explant, in a basic culture medium without plant growth hormones, adding 1.0-9.0 g/L of activated carbon, culturing for 5-21 d in dark and under scattered light for 16 h or 24 h daily and with the temperature of a culture room of 26-28 DEG C, carrying out in-vitro induction to obtain tracheids. The method for sorghum sudanense in-vitro induction of tracheid differentiation established by the invention has the tracheid differentiation rate reaching the highest value of 47.23%, and has the advantages that the method is simple and convenient in operation and high in induction efficiency, and provides a technical support for sorghum sudanense secondary cell wall lignin regulatory mechanism study and directional improvement.
