Abstract

The invention discloses a method for realizing tendon tissue regeneration by adopting stem cells from the periodontal membrane and the gum. The method is characterized in that a unique tendon tissue engineering 3d cell skeleton is provided by adopting a method that RGD tripeptide-coupled alginate microspheres are loaded with TGF-beta3, so that the microenvironment for differentiation of the odontogenic cells into the tendon tissue is improved, and meanwhile, the capability of the stem cells (extracted from the periodontal membrane and the gum) which are compounded with the alginate microspheres in participating in tendon regeneration is also evaluated. The in-vitro and in-vivo tests prove that the composite system can support the activity of the stem cells and differentiation into tendon-like tissues. The established odontogenic MSC serves as a seed cell for regeneration of the tendon tissue, and the micro-environment and an induction signal (TGF-beta3) play an important role in the activity of the odontogenic MSC and the differentiation towards the tendon-like tissue. The method has a good application prospect for optimizing tendon regeneration and repairing the limb bones.
