23 Jul Method for producing yeast expressed human tissue plasminogen activator and its activity detection
Abstract
The present invention relates to yeast expressed human tissue plasminogen activator (htPA) and the method of detecting its activity. On the basis of obtaining htPA cDNA, the 5′ end of the gene is modified to make it enter to the downstream of alpha factor signal sequence is pPIC9 vector. By the processes of engineering bacterium fermentation, induced expression, protein separation and purification and activity detection, pure rhtPA with molecular weight of 60 Kda is obtained. The experiments of immunity, enzymoimmunosoption, fibrin combination and degradation shows that the expression productis the same with natural histiotype plaminogen activator in performance.
