A preparation method method of biological sample embedding block based on nuclear forces microscope observation belongs to biological engineering. technical field, using stationary liquid of 2% pentanediol to low temperature fixation biological tissue sample or cell sample, which is then has rinsing by damping fluid of 0.2 mol/L ortho phosphoric acid and dehydration according to ethanol gradient of 50%->70%->94%-> absolute ethyl alcohol 10->ethanol then embedding after osmosis of ethoxyline and polymerization by heating with heating temperature and time of 35deg.C, 12 hours->45 deg.C, 12 hours->60 deg.C, 24 hours, at last acquiring biological sample embedding block suitable for embedding block microscope observation. The invention has fairly simple step, agent used is mostly produced in our country and easy to get with low cost, and is suitable for varies embedding block manufacture of animal tissue and cell sample.
