23 Jul Method for preparing universal fat-derived stem cells for medical tissue engineering
Abstract
The invention relates to a method for preparing universal fat-derived stem cells for medical tissue engineering, which comprises the following steps of: 1) cutting to obtain a fatty tissue; 2) digesting the fatty tissue obtained in the step 1 at the temperature of 37 DEG C for 40 to 60 minutes by using I-type collagenase and then separating to obtain primary fat-derived stem cells; and 3) constructing fusion expressed recombinant plasmids pEGFP-N-US2 and pEGFP-N-US3 for expressing US2 and US3 genes, transfecting the recombinant plasmids into the primary fat-derived stem cells by a liposome method, and performing screening and amplification culture to obtain the universal fat-derived stem cells for the medical tissue engineering. Due to the adoption of the method for preparing the universal fat-derived stem cells, the immunological rejection of heteroplastic transplantation is greatly reduced, and the universal fat-derived stem cells have good biological safety and lower immunogenicity, have universality, are applied to batch production of the stem cells in the tissue engineering, and are a novel material source for medical treatment.
