23 Jul Method for preparing tissue-engineering skin by utilizing skin derived progenitor cells (SKPs)
Abstract
The invention discloses a method for preparing tissue-engineering skin by utilizing skin derived precursors (SKPs). The method comprises the following steps: (1) separating SKPs; (2) transferring the separated SKPs into a multiplication culture medium for multiplying; (3) labeling SKPs with DiI fluorochrome; (4) preparing tissue-engineering skin, namely producing round collagen sponge with diameter of 8mm by a collagen sponge puncher, performing Co60 radiating sterilization, and adding SKPs cell 1*10<5>/20-30mu L culture medium to the rough surface of the sponge for culturing. The SKPs in the tissue-engineering skin can be differentiated into smooth muscle cells, vascular endothelial cells and nervous tissues, the differentiated smooth muscle cells and vascular endothelial cells can participate in formation of a blood vessel structure, and wound neovascularization is promoted, so that skin healing speed and healing quality can be improved.
