Abstract
The invention relates to a method for preparing a tissue engineering-oriented artificial tendon transplant based on an artificial tendon scaffold material. The method comprises the following steps: inoculating mesenchymal stem cells which are cultured to the third generation into a culture solution to culture; absorbing the culture solution, and adding a composite culture solution to infect for 4 hours; replenishing the composite culture solution to continually infect for 24 hours; absorbing the composite culture solution, and filling a culture solution containing 10 percent FBS (Fetal Bovine Serum) to infect continually for 20 hours to obtain cell sap carrying a report gene of GFP (Green Fluorescent Protein); transferring cells for expressing the GFP into the culture solution to culture in order to obtain seed cells carrying GFP-BMSCs; soaking an artificial tendon scaffold into a low-sugar DMEM (Dulbecco Modified Eagle Medium) to culture for 24 hours, planting GFP-BMSC cells onto the scaffold to culture continually for 48 hours, and transferring into tendon cell induced liquid to culture for 1-2 weeks in order to obtain an artificial tendon transplant. The prepared tissue engineering-oriented artificial tendon transplant can be planted into a human body to repair injured tendons.
