Abstract
The invention provides a method for preparing human definitive endoderm cells, and belongs to the technical field of stem cell induction and regeneration medical treatment. The method comprises the following steps: cloning a GDF3 gene to a lentiviral vector, and packaging a lentivirus; infecting human pluripotent stem cells with the packaged lentivirus, and performing resistance screening to obtain a stable expression cell line; and inducing the cell line to differentiate into the endoderm cells by using small molecules. The endoderm cells prepared by the method are generated through GDF3 induction, the cost is low, operation is easy, and the method can be used for preparing endoderm derived cells and applied to tissue engineering and has wide application prospects.
