Method for obtaining regenerated plants from hairy salvia miltiorrhiza roots

Abstract

The invention provides a method for obtaining regenerated plants from hairy salvia miltiorrhiza roots. The method comprises the following steps: cutting hairy salvia miltiorrhiza root explants newly generated under a dark culture condition into sections, placing in an induction culture medium, and performing dark culture for one month at 25 DEG C to induce formation of calluses; then transferring to a 25 DEG C and 16h/8h light/dark condition, culturing, and after about 7d, forming adventitious buds on the calluses; placing the calluses with the adventitious buds onto the induction culture medium, and continuing propagation culture; then cutting down the adventitious buds from the calluses, and transferring to a rooting culture medium, and culturing under a 25 DEG C and 16h/8h light/dark condition; when adventitious roots grow to 2-3 cm, transplanting to a greenhouse, and hardening seedlings to obtain salvia miltiorrhiza plants capable of growing normally. By the method for obtaining the regenerated plants from the hairy salvia miltiorrhiza roots, provided by the invention, the adventitious bud inducing rate is 50% or above and the rooting rate is 100%. The invention provides a novel tissue culture method for rapid propagation of the salvia miltiorrhiza plants and genetic engineering.
