Abstract
The invention discloses a method for obtaining an explant based on a heading-back twig in August to September to increase the survival rate of Dangshan crisp pear tissue culture seedlings. The method comprises the following steps: selecting a pear tree with a three age not exceeding 20 years in August to September, selecting an annual upward twig or a water sprout that grows well and does not suffer from plant diseases and insect pests, cutting the upward twig or the water sprout at a position where a bud is full and rich to induce the pear tree to germinate a new twig at the cutting wound, cutting the new twig as an explant for tissue culture when the new twig grows to 8-10cm, and subjecting the explant to sterilization, induction culture, proliferation and differentiation culture, scion culture, grafting and the like to obtain a Dangshan crisp crisp pear seedling. The method disclosed by the invention has the advantages of simplicity, easiness for operation and no addition of other costs; twigs can be treated in advance as required for the quantity of the explants based on a tissue culture plan. According to the method disclosed by the invention, sufficient regenerated Dangshan crisp pear plantlets can be obtained to meet the requirements for studying germplasm improvement or breeding Dangshan crisp pear seedlings on a large scale and the like by means of gene engineering; the survival rate can reach 96% or more based on the tissue culture method according to the invention.
