23 Jul Method for measuring in-vitro myocardial tissue contractility based on protein wire spring
Abstract
The invention discloses a method for measuring in-vitro myocardial tissue contractility based on a protein wire spring and belongs to the technical field of biomedical materials. Myocardial cells anda hydrogel solution are mixed and inoculated to areas of two parallel straight protein wires, after hydrogel gelatinization and myocardial cell contraction are started, the tensile length of a proteinwire spring is obtained through microscope observation, and the contraction force of the myocardial cells can be calculated according to a known elastic coefficient and Hooke’s law. The method for measuring the contractility of the in-vitro myocardial tissue based on the protein wire spring is simple, only winding and heating are needed, a complex processing technology is not needed, and a device(such as a PDMS micro-column) preparation method in the prior art is complex; the protein wire spring is high in precision, the elastic coefficient of the protein wire spring can reach 0.02 N/m, andthe myocardial cell beating contractility can be accurately measured; meanwhile, the preparation method is simple and convenient to operate and low in cost, and has a wide application prospect in thefield of tissue engineering.
