Abstract
The invention discloses a method for inducing bile duct epithelial cell differentiation in the construction of a tissue engineering bile duct unit taking ADSCs (Adipose Derived Stem Cells) as a foundation. The method comprises the following steps: A, obtaining adipose tissues from a target mammal; B, obtaining primary cells and carrying out subculture to obtain third-generation ADSCs; identifyinga surface marker of the third-generation ADSCs; C, preparing a polylactic acid block-shaped scaffold; preparing the third-generation ADSCs into a cell suspension solution and then slowly dropwise adding and covering a polylactic acid block-shaped scaffold material, and co-culturing; D, transplanting the polylactic acid block-shaped scaffold coated with the ADSCs into an abdominal cavity great omentum of the target mammal; E, injecting an EGF (Epidermal Growth Factor) and an HGF (Hepatocyte Growth Factor) to induce culture and differentiation; F, after inducing the culture and differentiation for 1 to 10 weeks, taking out the scaffold to obtain a bile duct epithelial cell of the target mammal.
