Abstract

The invention belongs to the technical field of plant genetic engineering, and discloses a method for increasing content of cytokinin of crop overground tissue. Through identification of rice OsABCG18as CK transport protein, an endogenous promoter sequence of OsABCG18 is used, and the overexpression of the OsABCG18 in rice is driven. Through quantifying of strains high in PCR screening expression, the content of CK in genetic modification strains is determined. The invention relates to the method which uses the endogenous promoter of the rice cytokinin transport protein OsABCG18 to drive theOsABCG18 gene expression to increase the content of cytokinin of crop overground tissue, and an application of the method. The content of the cytokinin of transgenesis plants is determined, and the economical character is analyzed, so that the cytokinin synthesized at roots can be proven to have the capacity for transportation to the overground tissue, the tiller amount and the panicles of the rice can be increased, and finally, the yield of rice grains can be increased.
