Abstract
The invention provides a method for establishing a callus induction and regeneration system for mature embryo of wild barley. The method comprises the following steps: 1) selecting wild barley seeds and disinfecting; 2) selecting an induction medium, a differentiation enrichment medium and a rooting culture medium; 3) treating and inducing differentiation of mature embryo: taking the wild barley seeds disinfected in step 1), putting into the induction medium for culturing, sub-culturing for several times and then transferring into the differentiation and enrichment medium for culturing, sub-culturing for several times and then transferring into the rooting culture medium for culturing, closing the bottle and hardening seedling when the roots of the differentiated seedlings are 4-6cm long, injecting running water after 2d, opening the bottle and hardening seedling for 3d, cleaning the roots, sterilizing and culturing with substrate, thereby acquiring a complete regenerated plant. The callus induction tissue culture method for wild barley, provided by the invention, is simple in operation, low in cost and unlimited by material taking time and space. An established regenerating method and system for mature embryo culture plant of wild barley can provide reliable technical support to the research on the seedling and genetic improvement of cell engineering.
