Abstract
The invention discloses a building method of adult middle-juncture goat skin epiderm stem cell system, which is characterized by the following: stripping 0.5 cm *0.5cm middle ear skin; cutting the skin into 0.1-0.2cm*0.1-0.2 cm tissue block; attaching skin in the glass or plastic plate with epiderm upwards; adding culture medium; culturing in the saturated dampness thermostatic incubator at 37 deg.c with 5 percent CO2; dispersing cultured skin into single cell; moving the single cell in the plastic plate through embryo moving pipe; culturing through non-serum culture medium; proceeding digestive passage culture through 0.2 percent pancreatic enzyme + 0.02 percent EDTA until the stem cell reaches 70-80 percent fusion; freezing large amount of seed cells to build the stem cell system of adult middle-juncture goat skin epiderm. The invention can be applied in cloning gene-transition animal and production, which provides ideal experience for tissue engineering study.
