23 Jul Method for direct quick amplification of quantitative mRNA with double chain combined with dye
Abstract
The invention is a method for directly and quickly amplifying quantitative mRNA level by using a double chain binding dye in the technical field of biological engineering, including the following steps: firstly, mRNA sequence to be detected is used for designing and synthesizing complementary probes; secondly, total RNAs of cells in thalli or tissue samples are extracted; thirdly, a specific probe is added for having protective hybridization with the target RNA; fourthly, S1 enzyme and RNase enzyme are added for eliminating single chain DNAs, unprotected RNAs, excessive probes and mismatched crossbreeds; and finally, a double chain binding dye is added for signal detection. The method is quick and easy, has strong specificity, high sensibility, good stability, good reproducibility and large application range, and is applicable to conventional laboratories.
