Method for building skin tissue based on biological 3D (three-dimensional) printing

Abstract

The invention discloses a method for building a skin tissue based on biological 3D (three-dimensional) printing. The method comprises the following steps of degreasing the skin tissue; using an acid-enzyme bonding method to treat, so as to obtain a supernatant; adding potassium carbonate, sodium carbonate, sodium chloride, potassium chloride, potassium sulfate or sodium sulfate into a crude extracting liquid, and salting out; using the obtained collagen as an ink raw material for printing the skin, filling into a printing chamber of a biological printer, and controlling the temperature of the printing chamber to 4 DEG C; using a laminating printing technique to print any shape of skin on a heat table with temperature of 37 DEG C; putting the printed skin into a container, and freeze-drying; putting the freeze-drying skin in formaldehyde or glutaraldehyde, crosslinking for 6h, and cleaning by a phosphate buffer solution; finally, soaking into an ethyl alcohol water solution, freeze-drying, and storing. The method has the advantages that the operation is simple and convenient; the skin restoration is effectively promoted; the beneficial reference is provided for the scientific research of artificial skin engineering, and clinical therapy of skin injury; a clinical effective method and technical support are provided for treating the diseases of skin wounds, burn wounds and the like.
