Abstract
The invention relates to the biotechnology field, and particularly discloses a method for activating expression of a granulocyte-macrophage colony-stimulating factor gene. The method comprises the following steps: 1) looking for a TALE target point sequence at an upstream of a promoter of the granulocyte-macrophage colony-stimulating factor gene, designing a target point recognition module for specific recognition of the TALE target point sequence, wherein the target point recognition module is composed of TAL nucleic acid recognition units; 2) constructing a target point recognition module coding sequence; 3) through an enzyme digestion connection method, connecting the target point recognition module coding sequence prepared in the step 2) with a skeleton carrier, to obtain a recombinant plasmid; and 4) transforming objective cells with the recombinant plasmid, and culturing. The TALE technology is utilized to activate expression of the granulocyte-macrophage colony-stimulating factor gene in autologous stem cells, a new cell source is provided for gene therapy of ischemic diseases, and an important role can be played in tissue engineering and cell therapy.
